Research programmes

Immunity (inflammation)

As AMC biobank and research ICT coordinator and head of AMC Biobank, I’m responsible for important parts of research support in the AMC, which are developed together with other university medical centers in the Netherlands. I represent the AMC within the main biobank infrastructures in the Netherlands – the Parelsnoer Institute and BBMRI. The ultimate goal for the coming years is the development of a national research infrastructure for disease prevention and personalized medicine: Health-RI.

In addition, I perform research in the field of immunology.

The role of adhesion G protein-coupled receptors in immunity
Since having cloned and deorphanized CD97 in the mid 90th, we are studying the biology of adhesion GPCRs:
• We discussed that their hybrid architecture is key to understanding the function and signaling of adhesion GPCRs (Sci Signal 2013)
• We confirmed the interaction of CD97 with its ligand CD55 in vivo obtained and obtained evidence for a role of mechanoperception in adhesion-GPCR biology (J Immunol 2013)
• Invited by the International Union of Basic and Clinical Pharmacology (IUPHAR), we developed a unified nomenclature for adhesion GPCRs (Pharm Rev 2015)
• We found that GPR56 expression is triggered by the transcriptional repressor Hobit, is downregulated upon cellular activation, and inhibits immediate effector functions in human NK cells (Cell Reports 2016)

The role of tissue-resident immune cells in inflammation
Aberrant immune activation is at the basis of various inflammatory diseases. We are investigating cellular programs in tissue-resident T cells and macrophages:
• We revealed that the human brain is populated by specialized tissue-resident memory T (TRM) cells with restricted cytotoxic capacity, possibly controlled by PD-1 (Acta Neuropathol 2013 and manuscript in preparation)
• We showed that chronic exposure to glucocorticoids shapes gene expression and modulates innate and adaptive activation pathways in macrophages with distinct changes in leukocyte attraction (J Immunol 2014)
• Studying gene expression in chronic active and inactive multiple sclerosis (MS) lesions, we identified genes that might be involved in the onset or regulation of demyelination, including scavenger receptors (manuscript in preparation)
• We developed a method for the immediate isolation and characterization of human microglia and obtained evidence that they persist in an active-prone state in MS (Glia 2012 & 2013 and Acta Neuropathol Commun 2017)
