WORKING EXPERIENCE

March '16-present:

Project manager (one day a week) for a new EU project (ESA-ITN, The European sepsis academy). Tasks; organize meetings, handle the website, organize the web-based recruitment for 15 new PhD positions (+ advertisement), organization of a postgraduate course and future tasks will also entail the writing of reports to the EU.

'09-present:

Post-doc/Project manager at the Amsterdam medical centre, department of experimental Immunology, Laboratory of Allergy, (Dr. R. Van Ree). Continuation of the work described below. In 2008 a new European project (FAST) started, aiming at the development of specific immunotherapy for food allergy. Within this project I handle the financial and scientific reporting to the EU, the toxicity studies and am part of the steering committee, which gives me new experience on the handling of large projects. In addition, in 2014 I also became the project manager for a new project, BM4SIT, which was launched to develop and improve immunotherapy for treatment of birch pollen allergy. In this project, as well as the FAST project (which started its second clinical trial in 2015) I am responsible for the organization of the clinical trials, writing the IMPDs, the METC application, the contact with the CRO and the Eudravigilance database registration.
 

'05-present:

Post-doc at the Amsterdam medical centre, department of experimental Immunology, Laboratory of Allergy, (Dr. R. Van Ree). Continuation of the work described below. In addition, a new programm (follow-up of the one described below), EuroPrevall, was started with 66 international partners. In the context of this project we work on isolating new emerging allergens from (tree)nuts and determining their prevalence. Within this project, I have extensive collaborations with many groups, representing the main centers working on allergy in Europe. In particularly, my work has focused on the isolation of oleosins, proteins present in the storage oilbodies of seeds and nuts and putatively new allergens leading to severe complaints. Also, we work on developing diagnostic tools to determine the levels of these allergens in foods. In addition my focus has been on determining the avidity/affinity of IgE antibody/antigen interactions and their importance for the subsequent clinical reactions.

’01-‘05

Post-doc at the Sanquin Foundation, department of Immunopathology (Dr. R. Van Ree). I worked in an international European project (SAFE) aimed at the determination of the geographical distribution of food allergies throughout Europe using apple allergy as a model system. 8 groups participated from 5 different countries. Part of this research involved a wide-ranging apple cultivar screening that required extensive collaboration between all participating researchers. In this project recombinant and natural proteins (apple allergens, Mal d 1-4) were produced/isolated and tested for their immune-reactivity with over 800 sera from different countries. Furthermore monoclonal and polyclonal antibodies were raised to the natural and recombinant proteins and assays were generated to detect the different allergens in vivo. In addition, for several specific patient sera, tests were done to determine the origin of their allergy.

PhD at the University of Leiden department of Plant Virology (Prof. Dr. J. F. Bol) in cooperation with 2 PhD’s at Wageningen Agricultural University:
Isolation and characterization of host factors (plasmodesmatal components?) interacting with the transport proteins (MPs) of Alfalfa mosaic virus (AMV) and cowpea mosaic virus (CPMV).
Using the two-hybrid system we tried to isolate plant host proteins interacting with the transport proteins of different plant viruses. After isolation these host proteins were further characterized and studied for their localization in the plant.

Research at the department of yeast genetics at the University of Leiden (Dr. H.Y. Steensma, Dr. A. Teunissen): Isolation and characterization of the FLO1 promoter in Saccharomyces cerevisiae.

TECHNIQUES

Computer programs, CAP, RAST(inhibition), Immunoblot(inhibition), RIA, ELISA, basophil histamine release, protein expression in yeast and bacteria. Work with a model system for histamine release in a rat basophilic leukemia cell-line (RBL) containing the alpha-chain of the human IgE receptor. Generating mouse monoclonals and purification of the monoclonal antibodies. Affinity and biochemical purification of several proteins (FPLC). Two-hybrid system, DNA, RNA and protein- isolations from bacterial-, yeast- and plant material, protein expression using different vectors, Immunoprecipitation techniques, Western, Southern and Northern blotting, in vitro transcription and translation, (RACE)-PCR, Agrobacterium tumefaciens agroinfiltration of plants, virus induced gene silencing (VIGS) and generating cDNA libraries. General Molecular biology techniques.

Internet FASTA/BLAST database searches, working with Microsoft office programs (word, excel, access, power point) and several in house data analysis programs.

ASSISTING/TEACHING TASKS

During my PhD and subsequent postdoctoral appointments I have supervised two technicians and 3 graduate students, in addition to several visiting PhD researchers from abroad. During my thesis I assisted several practical courses for first- to third year students, amongst which Molecular biology, Chemistry and Biochemistry courses. During my postdoctoral I have given a lecture for technicians in the department and assisted with a special course for first and second year medical students.
