Clinical Epigenomics: a Diagnostic Bridge Between Genotype and Phenotype in Mendelian Disorders

Dr. Bekim Sadikovic, PhD, DABMG, FACMG, is a Professor in the Department of Pathology in the Western University, Canada. He holds certifications in Clinical Molecular Genetics and Clinical Cytogenetics by the American Board of Medical Genetics. Professor Sadikovic serves as the Program Head of the Molecular Diagnostics Program in the Department of Pathology, the Director of the Verspeeten Clinical Genome Centre, and the Research Chair in Clinical Genomics and Epigenomics at the London Health Sciences, Canada.
Despite rapid advancements in genomic sequencing technologies, more than half of patients with genetic neurodevelopmental disorders remain without a molecular diagnosis. The key limitation is our ability to detect and interpret non-coding, complex and rare genetic variants. One molecular mechanism that is bridging the gap between the genotype and the disease phenotype involves epigenetic patterns of DNA methylation. The so-called DNA methylation “episignatures”, being a consequence to the underpinning genetic defect, represent a fundamental molecular “epi-phenotype”. Professor Sadikovic plays a pivotal role in the clinical adaption of episignature analysis as a sensitive and specific diagnostic modality for a rapidly expanding range of Mendelian disorders beyond standard genomic sequencing. Integration of genetic, epigenetic and clinical/phenotype databases is providing fundamental insights in the biology, complexity and expanding variety of human Mendelian conditions.
